Bracebrige OPP are investigating a fatal single vehicle crash in Georgian Bay Township.
The vehicle was travelling in the southbound lanes when it crashed between Lake Joesph Road and Twelve Mile Bay Road just before 7 Friday morning.
One woman was pronounced dead at the scene, two others have what is being described as life-altering injuries.
The driver was treated for minor injuries in hospital.
Southbound Highway 400 was closed at Lake Joseph Road for several hours but has since been reopened.
